From the Research
Daypro (oxaprozin) is a non-steroidal anti-inflammatory drug (NSAID) used to treat painful rheumatic and inflammatory conditions, offering the convenience of once-daily administration. It works by inhibiting cyclo-oxygenase, decreasing the formation of prostaglandin precursors from arachidonic acid, resulting in decreased prostaglandin biosynthesis and reduced pain and inflammatory responses 2. The medication is well absorbed after oral administration, with peak plasma concentrations reached in three to six hours, and is primarily eliminated by urinary excretion of the unchanged drug 2.
Key Characteristics of Daypro (Oxaprozin)
- Effective in treating rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, soft tissue disorders, and post-operative dental pain 3
- Offers strong analgesic qualities, particularly useful in painful musculoskeletal conditions such as periarthritis of the shoulder 3
- Has a high oral bioavailability of 95%, with peak plasma concentrations at 3 to 5 hours after dosing 3
- Metabolized in the liver by oxidative and conjugative pathways and readily eliminated by the renal and faecal routes 3
Dosage and Administration
- Anticipated daily dosage for adults is 600-1200 mg given as a single dose for rheumatoid arthritis, osteoarthritis, and analgesia 2
- In children, oxaprozin 10-20 mg/kg/day has been used to treat juvenile rheumatoid arthritis 2
Safety and Efficacy
- Oxaprozin has been shown to be as effective as other NSAIDs, such as aspirin, diclofenac, ibuprofen, and indomethacin, in several different painful musculoskeletal conditions 3
- Common side effects include gastrointestinal adverse effects, allergic reactions, analgesic nephropathy, hepatotoxicity, and increased bleeding times 2
- A prodrug form of oxaprozin has been developed to reduce gastrointestinal complications, showing significant analgesic, antipyretic, and anti-inflammatory activities with a remarkable decrease in the ulcer index 4
