Rybelsus (Semaglutide) Administration Guidelines
Rybelsus must be taken on an empty stomach with no more than 4 ounces (120 mL) of plain water, at least 30 minutes before the first food, beverage, or other oral medications of the day. 1

- GI side effects are typically mild to moderate and transient, resolving with continued treatment 1
- No dose adjustment is required for patients with renal impairment, including those with severe renal disease 1
Special Population Considerations
- For women using oral contraceptives, advise switching to a non-oral contraceptive method or adding a barrier method during Rybelsus initiation and dose escalation 1
- For patients planning to fast (e.g., during Ramadan), titrate to at least 7 mg approximately 2-3 weeks before the fasting period to ensure therapeutic efficacy and tolerance 1
- Monitor for hypoglycemia when used with insulin or insulin secretagogues (sulfonylureas) 1
Common Pitfalls to Avoid
- Taking Rybelsus with food or beverages other than water will significantly reduce absorption 1, 2
