From the Guidelines
Artane dosage should be 5 mg/kg once a day, with a usual dose of 300 mg for adults, as this has been shown to be effective in managing symptoms while minimizing side effects 1. When determining the appropriate dosage, it's essential to consider the patient's age and weight.
- For adults, the typical dosage is 300 mg, although doses of up to 450 mg may be used in some cases.
- For children between 12-17 years, the dosage is 5 mg/kg once a day.
- For children between 1 month and 11 years, the dosage is also 5 mg/kg once a day, although limited data is available for this age group 1. It's crucial to note that the dosage may need to be adjusted based on individual response and tolerance, and patients should be monitored for potential side effects.
- The medication should be taken orally, and the dosage can be adjusted as needed to achieve the desired therapeutic effect while minimizing adverse reactions 1.
From the FDA Drug Label
Idiopathic Parkinsonism As initial therapy for parkinsonism, 1 mg of trihexyphenidyl HCl in tablet form may be administered the first day. The dose may then be increased by 2 mg increments at intervals of three to five days, until a total of 6 to 10 mg is given daily The total daily dose will depend upon what is found to be the optimal level. Many patients derive maximum benefit from this daily total of 6 to 10 mg, but some patients, chiefly those in the postencephalitic group, may require a total daily dose of 12 to 15 mg Drug-Induced Parkinsonism The size and frequency of the trihexyphenidyl HCl dose needed to control extrapyramidal reactions to commonly employed tranquilizers, notably the phenothiazines, thioxanthenes, and butyrophenones, must be determined empirically The total daily dosage usually ranges between 5 and 15 mg although, in some cases, these reactions have been satisfactorily controlled with as little as 1 mg daily. Concomitant Use with Levodopa When trihexyphenidyl HCl is used concomitantly with levodopa, the usual dose of each may need to be reduced. Careful adjustment is necessary, depending on side effects and degree of symptom control. An trihexyphenidyl HCl dosage of 3 to 6 mg daily, in divided doses, is usually adequate Trihexyphenidyl HCl tablets - The total daily intake of trihexyphenidyl HCl tablets is tolerated best if divided into 3 doses and taken at mealtimes. High doses (>10 mg daily) may be divided into 4 parts, with 3 doses administered at mealtimes and the fourth at bedtime.
The recommended dosage of Artane (trihexyphenidyl) is:
- For idiopathic parkinsonism: 1 mg on the first day, increased by 2 mg every 3-5 days, up to 6-10 mg daily
- For drug-induced parkinsonism: 1-15 mg daily, determined empirically
- For concomitant use with levodopa: 3-6 mg daily, in divided doses The total daily dose should be divided into 2-4 doses, taken at mealtimes or bedtime, depending on the dose and patient tolerance 2
From the Research
Artane Dosage
- The dosage of Artane (trihexyphenidyl) is not explicitly stated in the provided studies, but its effects and pharmacokinetics are discussed in several studies 3, 4.
- In one study, patients with Parkinson's disease were administered 6 mg of trihexyphenidyl for 5 to 11 weeks, resulting in improvements in motor symptoms 4.
- The pharmacokinetics of trihexyphenidyl were studied in patients with dystonia, showing a biphasic semilogarithmic plot of serum concentration-time with a half-life of 3.7 +/- 0.4 hours 3.
- There is no direct information on the optimal dosage of Artane, but these studies provide insight into its effects and pharmacokinetics in patients with Parkinson's disease and dystonia 3, 4.
Related Studies
- A study comparing amantadine and trihexyphenidyl in the treatment of neuroleptic-induced parkinsonism found that both drugs were equally effective, but amantadine produced fewer side effects 5.
- Another study discussed the role of MAO-B inhibitors in managing fatigue in Parkinson's disease, but did not provide information on Artane dosage 6.
- A study on motor learning in Parkinson's disease highlighted the importance of understanding the processes involved in motor learning to provide effective medical and rehabilitative approaches, but did not mention Artane dosage 7.
