Nicotinamide mononucleotide (NMN) supplementation is generally well-tolerated, with most side effects being mild and transient, and the most recent and highest quality study suggests that oral administration of NMN is safe and effectively metabolized in healthy individuals without causing any significant deleterious effects 1. The potential side effects of NMN supplementation include:
- Nausea
- Stomach discomfort
- Diarrhea
- Headaches
- Fatigue
- Insomnia Some users may also experience skin flushing or rashes, similar to what occurs with niacin supplements. At higher doses (above 1000mg daily), more pronounced gastrointestinal issues may occur. NMN might also interact with medications, particularly those affecting blood sugar levels, as it can influence insulin sensitivity. There have been rare reports of increased anxiety or heart palpitations in sensitive individuals. According to the study published in the Endocrine Journal in 2020, single oral administrations of NMN did not cause any significant clinical symptoms or changes in heart rate, blood pressure, oxygen saturation, and body temperature 1. The study also found that plasma concentrations of N-methyl-2-pyridone-5-carboxamide and N-methyl-4-pyridone-5-carboxamide were significantly increased dose-dependently by NMN administration. Another study published in Clinical Nutrition ESPEN in 2023 found that oral administration of NMN elevated plasma levels of NMN and NAD+, and postprandial serum insulin levels, with no adverse symptoms observed in the participants 2. It is recommended to start with lower doses (250-500mg daily) and gradually increase if needed, as the body's response to NMN varies significantly between individuals due to differences in metabolism, age, and overall health status 1. If side effects persist or are severe, discontinuing supplementation and consulting a healthcare provider is recommended, especially for those with pre-existing health conditions or who are taking other medications.
